| Background: | The I-ï«B kinase complex (IKK) functions as a mediator of NF-ï«B activation in response to multIPPle stimuli by phosphorylating I-ï«B inhibitor proteins and causing their degradation. IKK contains two structurally related catalytic subunits termed IKKï¡ and IKKï¢ as well as a noncatalytic subunit called IKKï§/NEMO. IKK is composed of an IKKï§/NEMO homodimer bound together with either an IKKï¡/IKKï¢ heterodimer or an IKKï¢ homodimer Although IKKï§/ NEMO itself does not have kinase activity, it is essential for NF-ï«B activation, most likely via a scaffold function that is required for IKK activity. Mutations in the X-linked gene encoding IKKï§ can interfere with NF-ï«B signaling and lead to immunodeficiency disease. |
