Background:The I-B kinase complex (IKK) functions as a mediator of NF-B activation in response to multIPPle stimuli by phosphorylating I-B inhibitor proteins and causing their degradation. IKK contains two structurally related catalytic subunits termed IKK and IKK as well as a noncatalytic subunit called IKK/NEMO. IKK is composed of an IKK/NEMO homodimer bound together with either an IKK/IKK heterodimer or an IKK homodimer Although IKK/ NEMO itself does not have kinase activity, it is essential for NF-B activation, most likely via a scaffold function that is required for IKK activity. Mutations in the X-linked gene encoding IKK can interfere with NF-B signaling and lead to immunodeficiency disease.
