| Background: | Kaiso is a member of the POZ-zinc finger family of transcription factors implicated in development and cancer. It acts as a DNA methylation-dependent transcriptional repressor. Kaiso is a dual specificity DNA-binding protein that recognizes the specific consensus sequence CTGCNA as well as methyl-CpG dinucleotides. Two copies of the Kaiso-binding site are present in the matrilysin promoter, implicating matrilysin as a candidate target gene for Kaiso. Kaiso also associates with the adhesion molecule p120ctn and may be a downstream effector of cadherin and/or p120ctn signaling. The DNA-binding ability of Kaiso with either recognition site was inhibited by p120ctn. Thus, Kaiso may link events at the cell surface with DNA methylation-dependent gene silencing. |
