Background: 

Cdk4 (Cyclin-Dependent Kinase 4) is a 34 kDa kinase involved in the control of cell proliferation and progression between the G1 and S phases of the cell cycle. Cdk4 acts as a G1 checkpoint; it is constitutively expressed and the assembly of cyclin D/Cdk4 complexes is the rate-limiting step in progression through the G1 stage of the cell cycle. p16, a putative tumor suppressor, also plays a role in cell cycle regulation by binding to and inhibiting Cdk4 in competition with cyclin D. Upregulation of the cyclin D/Cdk4/6 complex is associated with the early stages of neuronal apoptosis, and the cyclin D/Cdk4 complex phosphorylates pRB (retinoblastoma gene product). Since p16 and pRB activity are inactivated or modified in a wide variety of human cancers, Cdk inhibitors such as Cdk4 may have therapeutic potential as anti-cancer drugs.
