| Background: | When T-cells and NK-cells recognize target cells, the contents of the lytic granules, including proteins such as perforin, granzymes and granulysin, are secreted by degranulation. CD107a, also known as lysosome-associated membrane protein 1 (LAMP-1), is a glycoprotein present in the membrane of cytotoxic granules. As a result of degradation by these lytic molecules, CD107 is exposed on the cell surface. Thus surface-exposed CD107 is a marker of secretion of the lytic granule contents, and the detection of CD107 exposed on the cell surface enables rapid assessment of cell-mediated cytotoxicity and detection of antigen specific cytotoxic T-cells. The “CD107a mobilization assay†is the most commonly used method for detection of CD107a. In this method, the cells are cultured in the presence of CD107a monoclonal antibody after antigen stimulation, which enables sensitive detection of CD107a.
